Effects of <i>Buyang Huanwu</i> Decoction on Endoplasmic Reticulum Stress-Autophagy Response and GADD34 Expression in Mice with CIRI

Objective: To investigate the effects of Buyang Huanwu Decoction on endoplasmic reticulum stress-autophagic response and expression GADD34 in ischemic penumbra mice with CIRI. Methods: The were randomly divided into sham operation group, model group low-, medium-, high-dosage groups. treated by gavage for 7 days before modeling 24 hours after modeling. CIRI was established thread embolization method. neurological function evaluated 48 modeling; TTC staining used to observe cerebral infarct volume; HE pathological changes penumbra; TUNEL performed assess cell apoptosis; Western blot stress marker protein GRP78 autophagy proteins LC3 p62; Immunohistochemistry real-time PCR applied examine mRNA expressions GADD34. Results: Compared score volume infarction decreased, reperfusion injury improved varying degrees, number apoptotic cells p62 down-regulated while ratio LC3-II/ –I up-regulated, up-regulated each statistical significance (P<0.01). Conclusion Buyang can significantly improve damaged function, histopathological damage penumbra, as well reduce apoptosis cells, which plays a neuroprotective role Its mechanism may be related up regulating GADD34, steady-state balance penumbra.
